MY can be M' as in M'LUD, M'LORD
m is an abbreviation for minutes, in such as rpm, revolutions per minute,

Ha! Having posted an hour ago that M is in Chambers for Minute, I'd failed to spot, a couple of entries further on, that M' is an abbreviation for MY.

Dsm101 is absolutely correct to say that the preamble is inaccurate in relation to the resolution of ambiguities. It didn't cause me a problem because I surmised, as everyone else has done, that the final grid consists of real words, but that doesn't alter the fact that the wording of the preamble is wrong in relation to 6a and 44a.

Presumably the setter didn't want to reveal prematurely that the final grid consists of real words. All that was needed was a minor caveat, and maybe a reference to the puzzle's title to resolve the last few ambiguities.

It's row as a verb, to disagree with, quarrel etc.
A (one) + 3 letter shrub, with G in it.

Hello all
Hope I've not missed it, but in the across answers, if letters do not need to be changed, do they stay in the same position.
Would that mean that 40A is ??N?

Yes that is correct.

That is how you can clear up ambiguities in final grid. The entries have to be real words. So without knowing the original across answer and jumble you would have to guess between midi and mini in the final grid.
