20d from the bible - parable of the wedding at ......water turned to wine , then put a T in that answer, then change the 1st 4 letters to its opposite/positive

4dn not Japanese, a straight clue, think of throw like confuse. I also came late to this, a hard one. My problem was, because I jump to answer without completely solving wordplay, it took me ages to realise that the word play was for what went in the grid, not for the definition. Done now, time for the Speccie.

I have 4d, Teenieleek. It is 5d that I'm not sure about. I have ?O?O ?Y?S, which I think has a 'Japanese' connection.

I no longer have my copy, but there's definitely a Japanese connection for 5 D. The 'y' was an 'n' before any change was made.
