I thought sleeping on it might bring inspiration this morning but alas I am still stuck on 12d, bit by bt, 45d, grappling hook, and 56a, merciful/tolerant. Can someone help please? I can't start my day until I've sorted these!

43d took on could also be "appointed" as in appointing someone to a job, I.e. you take them on.

Thanks Jazzgirl, so 44a is "depressing" , not "depression". I usually find that when I am really stuck its because I've made a mistake.
